Pharmacologic action:

Pharmacokinetics:

Safety and effectiveness:
The pharmacokinetics of vardenafil, dapoxetine, and their metabolites have been studied in 24 healthy male volunteers. They were given orally one time each of vardenafil 10 mg, 60 mg, and 100 mg (three times daily). The volunteers were divided into three groups:
- Group A (n=6) - Dose was increased to 5 mg/kg every other day for 1 day (40 mg/day), to 6 mg/kg every other day for 3 days (40 mg/day), and to 10 mg/kg every other day for 4 days (100 mg/day).
Group B (n=6) - Dose was increased to 10 mg/kg every other day for 1 day (40 mg/day), to 6 mg/kg every other day for 3 days (40 mg/day), and to 10 mg/kg every other day for 4 days (100 mg/day).
Group C (n=6) - Dose was increased to 10 mg/kg every other day for 1 day (40 mg/day), to 6 mg/kg every other day for 3 days (40 mg/day), and to 10 mg/kg every other day for 4 days (100 mg/day).
In this study, the mean plasma vardenafil concentration was 0.08 mg/L (range 0.02 to 0.22 mg/L). Mean oral doses of vardenafil and dapoxetine were 7.1 and 18.3 mg, respectively.
